dc.description.abstractBovine herpesvirus type 5 (BoHV-5) is an alphaherpesvirus associated with meningoencephalitis, an important disease of cattle in South America. Alphaherpesvirus recombinants have been used to study the function of gene products and also for vaccine production. In particular, the gene encoding the enzyme thymidine kinase (tk) has been deleted to produce attenuated strains. Thymidine kinase is required for efficient replication of these viruses in the central nervous system (CNS), and its deletion or inactivation is associated with reduction of virulence. Thus, the present study aimed to investigate the pathogenesis and to characterize clinically and virologically the acute and latent infection by BoHV-5 (strain SV-507/99) and by tk-deleted recombinant (BoHV-5tkΔ) in lambs. Chapter 1 reports the characterization of latent by BoHV-5 infection in lambs. During acute infection, lambs inoculated with BoHV-5 (SV-507/99) shed virus in nasal secretions during up to 11 days and developed viral neutralizing antibodies in titers ranging from 16 to 128. Administration of corticosteroids during latency (day 65 pi) resulted in virus reactivation and shedding for 3 to 8 days in titers up to 64. Thus, it is proposed that sheep can be used as animal models for studying the biology of latent infection by BoHV-5. In Chapter 2, was investigated the ability of BoHV-5tkΔ to establish and reactivate latent infection in lambs. After viral inoculation, the recombinant virus retained the ability to replicate in the nose of the lambs. At day 40 pi, latent viral DNA was detected in the trigeminal ganglia (TG) in all inoculated animals. However real time PCR demonstrated a significantly reduced amount of recombinant DNA (9.7-fold less, p<0.001) compared to the parental virus. Following dexamethasone (Dx) treatment, infectious virus was not detected in nasal secretions of the animals inoculated with the recombinant virus, contrasting with virus shedding by lambs inoculated with the parental virus. Nevertheless, some nasal swabs from animals inoculated with BoHV-5tkΔ were positive for viral DNA, indicating low levels of reactivation. Thus, BoHV-5 TK activity is not required for the establishment of latency, but seems critical for efficient virus reactivation. Chapter 3, reports the ability of BoHV-5 and BoHV-5tkΔ to establish and reactivate latent infection in lymphoid tissues. During latent infection, viral DNA was detected in TGs, pharyngeal and palatine tonsils and in retropharyngeal lymph node of all sheep inoculated with both viruses (parental and recombinant). At day 40 pi, Dx was administered to the animals to induce viral reactivation. Viral mRNA of BoHV-5 glycoprotein B was detected in TGs, retropharyngeal lymph node and tonsils of lambs inoculated with the parental virus. In animals inoculated with BoHV-5tkΔ, gB mRNA was detected only in tonsils. Therefore, it is suggested that lymphoid tissues are also sites of latency by BoHV-5 and the recombinant BoHV-5tkΔ, and that viruses can be reactivated from these sites. However, reactivation of recombinant virus appears to occur only in lymphoid tissues and not in neural tissues. In summary, the presented results contribute for the understanding of the pathogenesis and characterization of infection by BoHV-5 and BoHV-5tkΔ.eng

dc.description.resumoO herpesvírus bovino tipo 5 (BoHV-5) é um alfaherpesvírus associado com meningoencefalite, uma doença de grande importância em bovinos na América do Sul. Recombinantes de alfaherpesvírus defectivos em genes não essenciais vem sendo utilizados para o estudo da função de produtos gênicos e também para a produção de vacinas. O gene da enzima timidina quinase (tk) tem sido deletado para produzir cepas atenuadas com fins vacinais. A enzima timidina quinase (TK) é necessária para a replicação eficiente desses vírus no sistema nervoso central (SNC), e a sua deleção ou inativação está associada com a redução da virulência. Assim, o presente trabalho teve como objetivo estudar a patogenia e caracterizar aspectos clínicos e virológicos da infecção aguda e latente pelo BoHV-5 (cepa SV-507/99) e por um recombinante com deleção no gene da tk (BoHV-5tkΔ) em ovinos. O capítulo 1 relata a caracterização da infecção pelo BoHV-5 em ovinos. Durante a infecção aguda, cordeiros inoculados com o BoHV-5 (SV-507/99), excretaram vírus em secreções nasais por até 11 dias pós inoculação (pi) e desenvolveram anticorpos neutralizantes em títulos de 16 até 128. A administração de corticoides durante a latência (dia 65 pi) resultou em reativação e excreção viral durante 3 a 8 dias. Sugere-se, assim, que ovinos podem ser utilizados como modelos animais para o estudo da biologia da infecção latente pelo BoHV-5. No capítulo 2, investigou-se a capacidade do BoHV-5tkΔ em estabelecer e reativar a infecção latente em ovinos. Após inoculação viral, o vírus recombinante manteve a capacidade de replicar na cavidade nasal. No dia 40 pi, DNA viral latente foi detectado nos gânglios trigêmeos (TG) de todos os animais, porém demonstrou-se, uma significativa redução (9,7 vezes; p<0,001) na quantidade de DNA do vírus recombinante em relação ao vírus parental, determinado por PCR em tempo real. Após tratamento com dexametasona (Dx), não foi detectado vírus nas secreções nasais dos animais inoculados com o vírus recombinante, contrastando com a excreção viral nos ovinos inoculados com o vírus parental. Porém, alguns suabes nasais dos animais inoculados com o BoHV-5tkΔ foram positivos para o DNA viral, indicando assim, baixos níveis de reativação. Logo, a atividade da TK no BoHV-5 não é necessária para o estabelecimento de latência, mas é crucial para uma reativação eficiente. O capítulo 3, relata uma investigação da capacidade do BoHV-5 e BoHV-5tkΔ em estabelecer e reativar a infecção latente em tecidos linfoides. Durante infecção latente, DNA viral foi detectado nos TGs, tonsilas faringeal e palatina e no linfonodo retrofaríngeo de todos ovinos inoculados com ambos os vírus (parental e recombinante). No dia 40 pi, foi administrada Dx nos animais para induzir reativação viral. Foi detectado RNA mensageiro (mRNA) da glicoproteína B (gB) do BoHV-5, no TG, linfonodos retrofaríngeos e tonsilas dos cordeiros inoculados com o vírus parental. Nos animais inoculados com o BoHV-5tkΔ, mRNA foi detectado apenas nas tonsilas. Logo, sugere-se que tecidos linfoides também são sítios de latência para o BoHV-5 e do recombinante BoHV-5tkΔ, podendo estes vírus serem reativados à partir destes locais. A reativação do recombinante, no entanto parece ocorrer apenas em tecidos linfoides e não em tecidos neurais. Em resumo, os resultados deste estudo contribuem para o conhecimento da patogenia e caracterização da infecção pelo BoHV-5 e pelo recombinante.por
