dc.description.abstract | The richness of Brazilian biodiversity and the popular tradition of plants known as medicinal make phytotherapy widely used today. Among the plants used, the species Leandra australis, popularly known as pixirica, stands out. This species is popularly used for inflammatory diseases and as a weight-loss. Despite its popular uses, there are few studies in the current literature on its chemical molecules and pharmacological activities. Furthermore, so far, no toxicological studies have been found with this plant species. Therefore, this work aimed to evaluate the chemical composition, toxicity and antioxidant activity of L. australis. The plant was collected in March/2015, identified and extracted by maceration, with etanol 50%. Subsequently, its crude extract was fractionated with increasing polarity solvents (hexane, chloroform, ethyl acetate and butanol). The crude extract, fractions and remaining aqueous residue of the fractionation were concentrated in a rotary evaporator at a temperature below 45°C. The evaluation of the chemical composition by High Performance Liquid Chromatograph (HPLC) coupled to Mass Spectrometry was performed with the crude extract. Quantifications of secondary metabolites (total polyphenols, flavonoids, condensed tannins and total alkaloids) were performed in Spectrophotometry UV-Vis with the crude extract, fractions and aqueous residue. The evaluation of the antioxidant activity of the extract, fractions and aqueous residue was carried out by three different methods also carried out in Spectrophotometer UV-Vis, namely: evaluation of DPPH radical capture, determination of iron reduction ability (FRAP), and co-oxidation with beta-carotene and linolenic acid. The evaluation of the toxicity of the crude extract was carried out using alternative methods in vivo, being the cytotoxicity carried out on Artemia salina and the genotoxicity carried out on Allium cepa radicle cells. Toxicity was also assessed from the OECD 423 and 407 acute toxicity and repeated dose 28-day protocols using Wistar rats. The samples with the highest yield were aqueous residue, butanol and crude extract. In the assays, the high content of phenolic compounds and condensed tannins in the samples stood out, being also possible to quantify the gallic acid (19.32±0.39 mg/g of extract), rutin (12.30±0.01 mg/ g of extract) and quercetin (162.58 ± 0.68 mg/g of extract) in the plant extract analyzed in HPLC. The species under study showed considerable antioxidant activity in its extract and fractions. The toxicity profile of the species, unprecedented, did not show cytotoxicity to A. salina (LC50 1607.15 μg/mL), but a possible antiproliferative (genotoxic) activity in the crude extract of this species was evidenced. The evaluation of toxicity in Wistar rats showed that the acute administration and repeated doses did not cause deaths or behavioral changes in the animals. However, acute administration identified changes in BUN levels in the animals tested, which shows possible kidney damage. When administered continuously for 28 days, the plant did not cause hematological changes and the biochemical evaluation did not identify signs of toxicity. These results show that the plant species has low toxicity and reinforces the importance of conducting studies on chronic oral administration in order to confirm its safety for medicinal use. | eng |

dc.description.resumo | A riqueza da biodiversidade brasileira e a tradição popular sobre plantas ditas como medicinais faz com que a fitoterapia seja amplamente utilizada nos dias atuais. Dentre as plantas utilizadas, destaca-se a espécie Leandra australis, conhecida popularmente como pixirica. Esta espécie é utilizada popularmente para doenças inflamatórias e como emagrecedora. Apesar de seus usos populares, há poucos estudos na literatura atual sobre suas moléculas químicas e atividades farmacológicas. Além disso, até o momento, não foram encontrados estudos toxicológicos com esta espécie vegetal. Sendo assim, este trabalho teve como objetivo avaliar a composição química, toxicidade e atividade antioxidante de L. australis. A planta foi coletada em Março/2015, identificada e extraída por maceração, com etanol 50%. Posteriormente, seu extrato bruto foi fracionado com solventes de polaridade crescente (hexano, clorofórmio, acetato de etila e butanol). O extrato bruto, frações e resíduo aquoso restante do fracionamento foram concentrados em evaporador rotatório sob temperatura inferior a 45°C. A avaliação da composição química por Cromatógrafo Líquido de Alta Eficiência (CLAE) acoplado à Espectrometria de Massas foi realizada com o extrato bruto. As quantificações de metabólitos secundários (polifenóis totais, flavonoides, taninos condensados e alcaloides totais) foram realizadas em Espectrofotometria UV-Vis com o extrato bruto, frações e resíduo aquoso. A avaliação da atividade antioxidante do extrato, frações e resíduo aquoso ocorreu por três métodos diferentes realizados também em Espectrofotômetro UV-Vis, sendo eles: avaliação da captura do radical DPPH, determinação da habilidade de redução do ferro (FRAP), e co-oxidação com betacaroteno e ácido linolênico. A avaliação da toxicidade do extrato bruto foi realizada a partir de métodos alternativos in vivo, sendo a citotoxicidade realizada em Artemia salina e a genotoxicidade realizada em células de radículas de Allium cepa. A toxicidade também foi avaliada a partir de protocolos de toxicidade aguda e de doses repetidas de 28 dias da OECD 423 e 407 utilizando ratos Wistar. As amostras com maior rendimento foram resíduo aquoso, butanol e extrato bruto. Nos doseamentos destacaram-se o elevado teor de compostos fenólicos e taninos condensados nas amostras, sendo também possível quantificar o ácido gálico (19,32±0,39 mg/g de extrato), rutina (12,30±0,01 mg/g de extrato) e quercetina (162,58±0,68 mg/g de extrato) no extrato da planta analisado em CLAE. A espécie em estudo apresentou considerável atividade antioxidante em seu extrato e frações. O perfil de toxicidade da espécie, inédito, não apresentou citotoxidade para A. salina (CL50 1.607,15 μg/mL), porém foi evidenciada uma possível atividade antiproliferativa (genotóxica) no extrato bruto desta espécie. A avaliação da toxicidade em ratos Wistar mostrou que a administração aguda e de doses repetidas não ocasionou mortes ou alterações comportamentais nos animais. Porém, a administração aguda identificou alterações nos níveis de BUN nos animais testados, o que mostra possível dano renal. Quando administrada de forma contínua por 28 dias, a planta não ocasionou alterações hematológicas e a avaliação bioquímica não identificou sinais de toxicidade. Estes resultados mostram que a espécie vegetal apresenta baixa toxicidade e reforça a importância da realização de estudos sobre administração oral crônica a fim de confirmar a segurança para seu uso medicinal. | por |
