dc.description.abstract | The influenza B virus, the causative agent of influenza, is still one of the main causes of
respiratory diseases that affects the world's population and causes thousands of deaths
worldwide. Oxidative stress, may be associated with this disease, caused by alterations in the
production of reactive oxygen species (ROS) and the imbalance of antioxidants, which leads to
the oxidation of biomolecules and loss of their biological functions. The sulfhydryl enzyme
delta-aminolevulinate dehydratase (δ-ALA-D) may be inhibited in pro-oxidant situations and
this inhibition also causes an increase in the production of free radicals. Thus, the δ-ALA-D
enzyme may be an indirect marker of oxidative stress. Furthermore, there is the production of
pro-inflammatory cytokines, which, in excess, can also result in injuries to the infected
organism and result in cell and tissue damage and consequent serious conditions. In this context,
considering the thousands of infections and deaths from influenza B every year, the objective
is to verify the oxidative and inflammatory profile and the activity of the δ-ALA-D enzyme in
the infection by the influenza B virus. Blood samples from 50 participants were evaluated as
influenza B virus and 30 healthy participants. The oxidative stress quantified in the analyzed
groups were through thiobarbituric acid reactive substances (TBARS), protein (PSH) and nonprotein (NPSH) thiol groups, vitamin C levels (VIT C), plasma iron reduction ability (FRAP)
and the enzyme delta-aminolevulinate dehydratase (δ-ALA-D). The inflammatory profile was
analyzed by determining the levels of interleukin-2 (IL-2), interleukin-4 (IL-4), interleukin-6
(IL-6), interleukin-10 (IL-10), interleukin-17A (IL-17A), interferon-gamma (IFN-γ) and tumor
necrosis factor alpha (TNF-α). The results showed that lipid peroxidation and levels of IL-6,
IL-10, IL-17A and IFN-y were statistically higher in the group with influenza B. The activity
of analyzed antioxidants, levels of thiols, vitamin C and FRAP, was significantly lower in those
infected with the influenza B virus compared to the controls, as well as the activity of the δALA-D enzyme, in addition the enzyme showed a higher reactivation rate for those infected
with influenza B. generation of oxidants, the depletion of the antioxidant system, reduced
activity of the δ-ALA-D enzyme and an expression pattern of IL-6, IL-10, IL-17A and IFN-y
during influenza B virus infection. | eng |

dc.description.resumo | O vírus da influenza B, agente causador da gripe, é ainda uma das principais causas de doenças
respiratórias que acomete a população mundial e causador de milhares de mortes no mundo. O
estresse oxidativo, pode estar associado a essa doença, causado por alterações na produção de
espécies reativas de oxigênio (EROs) e no desequilíbrio de antioxidantes, que leva à oxidação
de biomoléculas e perda de suas funções biológicas. A enzima sulfidrila delta-aminolevulinatodesidratase (δ-ALA-D) pode estar inibida em situações pró-oxidantes e essa inibição, ainda,
causa um aumento na produção de radicais livres. Assim, a enzima δ-ALA-D pode ser um
marcador indireto de estresse oxidativo. Ainda, tem-se a produção de citocinas próinflamatórias, que, em excesso, pode também resultar em agravos no organismo infectado e
resultar em danos nas células e nos tecidos e consequente quadros graves. Nesse contexto,
considerando as milhares de infecções e mortes por influenza B todos os anos, tem-se como
objetivo verificar o perfil oxidativo e inflamatório na infecção pelo vírus da influenza B. Foram
avaliadas amostras sanguíneas de 50 participantes com o vírus da influenza B e 30 participantes
saudáveis. O estresse oxidativo foi quantificado nos grupos analisados através das substâncias
reativas ao ácido tiobarbitúrico (TBARS), como medida da peroxidação lipídica, dos
antioxidantes como grupos tióis protéicos (PSH) e não protéicos (NPSH), níveis de vitamina C
(VIT C), habilidade plasmática de redução do ferro (FRAP) e por fim da enzima deltaaminolevulinato-desidratase (δ-ALA-D). O perfil inflamatório foi analisado através da
determinação dos níveis de interleucina-2 (IL-2), interleucina-4 (IL-4), interleucina-6 (IL-6),
interleucina-10 (IL-10), interleucina-17A (IL-17A), interferon-gama (IFN-γ) e fator de necrose
tumoral alfa (TNF-α). Os resultados demonstraram que a peroxidação lipídica e os níveis de
IL-6, IL-10, IL-17A e IFN-y foram estatisticamente maiores no grupo com influenza B. A
atividade dos antioxidantes analisados, níveis de tióis, vitamina C e FRAP, foi
significativamente menor em infectados com o vírus do influenza B em comparação com os
controles, assim como a atividade da enzima δ-ALA-D. Além disso, essa enzima apresentou
maior índice de reativação para os infectado com influenza B. Dessa forma, evidenciamos a
geração de oxidantes, a depleção do sistema antioxidante, a atividade reduzida da enzima δALA-D e um padrão de expressão de IL-6, IL-10, IL-17A e IFN-y durante a infecção pelo vírus
