dc.description.abstract | Drosophila melanogaster is an model organism that have been used to research molecular mechanisms and role of the genes and proteins involved in nociception, which have a function in detect noxious stimuli and protect the tissues of an potential damage. Molecular mechanisms that act in detecting and conducting of the nociceptive signal, involving ions channels as Transient Receptors Potential (TRP) and neurotransmitters. In D. melanogaster, TRPA1 is a receptor have activated by heat, participating in the induction of thermal nociceptive behavior as avoidance of noxious heat. The monoamines have been involved on physiological and behavioral process, for example the dopamine that supports in the control of locomotor activity and temperature sensitivity. However, little is known about the participation of serotonin in behavioral responses to heat. Therefore, we investigated whether the monoamines participate in the neuronal communication of the nociceptive pathway for the processing and response to thermal nociceptive stimuli. In this study, we research the effects of the depletion of monoamines and, selectively, oh the dopamine e serotonin, through of pharmacological manipulation, on thermal nociceptive behaviors in the D. melanogaster. Flies were treated with reserpine (10 μM, 30 μM, 100 μM and 300 μM), 3-Iodo-L-Tyrosine (5 mg and 10 mg) and p-chloro-phenylalanine (5 mg and 10 mg), then, it was subjected to behavioral tests. The reserpine reduces the thermal tolerance profile and, thus, the heat resistance of the D. melanogaster, as well as the avoidance of noxious heat. Whereas treated flies have a deficit locomotion induced by reserpine, there are flies able to execute the avoidance behavior. Also, PCPA decrease avoidance of noxious heat and heat resistance at 41º C without have impaired locomotion, suggesting that the impaired avoidance of the noxious heat is related to the reduction of the heat resistance in the D. melanogaster by this drugs. These data suggest that monoamines, particularly serotonin, are associated to the impaired avoidance of the noxious heat which could be related to the reduction of the heat resistance in the D. melanogaster. | eng |

dc.description.resumo | A Drosophila melanogaster é um organismo-modelo usado para estudar mecanismos moleculares e funções dos genes e proteínas envolvidas na nocicepção. A dor nociceptiva tem a função de detectar estímulos nocivos aos tecidos e, assim, protegê-los de potenciais danos. Os mecanismos moleculares que atuam na detecção e condução do sinal nociceptivo envolvem canais iônicos, como os receptores de potencial transitório (TRP), e os neurotransmissores. Nas D. melanogaster o TRPA1 é um receptor ativado pelo calor e participa na indução de comportamentos nociceptivos térmicos como a esquiva ao calor nocivo. As monoaminas estão envolvidas em processos fisiológicos e comportamentais, como a dopamina que auxilia no controle da atividade locomotora e sensibilidade térmica. No entanto, pouco se sabe sobre a participação da serotonina em respostas comportamentais ao calor. Dessa forma, nós investigamos se as monoaminas participam na comunicação neuronal da via nociceptiva para o processamento e resposta aos estímulos nociceptivos térmicos. Neste estudo, investigou-se o envolvimento da depleção de monoaminas e, seletivamente, da dopamina e serotonina, através da manipulação farmacológica, sobre os comportamentos de nocicepção térmica de D. melanogaster. Para isso, as moscas foram tratadas com reserpina (10 μM, 30 μM, 100 μM e 300 μM), 3-Iodo-L-Tirosina (5 mg/mL e 10 mg/mL) e p-clorofenilalanina (5 mg/mL e 10 mg/mL), em seguida, foram submetidas aos testes comportamentais. A reserpina reduziu o perfil de tolerância térmica, ou seja, a resistência ao calor da D. melanogaster, bem como o comportamento da esquiva ao calor nocivo. Apesar de possuir um déficit locomotor induzido pela reserpina, haviam moscas que foram capazes de executar o comportamento de esquiva Além da reserpina, o PCPA diminui a esquiva ao calor nocivo e a resistência ao calor à 41° C sem apresentar dano locomotor, sugerindo que a perda da habilidade de esquiva ao calor nocivo esteja relacionada com a diminuição da resistência ao calor em D. melanogaster por estes fármacos. Os dados do presente estudo sugerem que as monoaminas, particularmente a serotonina, estão associadas ao prejuízo na esquiva ao calor nocivo o qual pode estar relacionado à redução da resistência ao calor em D. melanogaster. | por |
