dc.description.abstract | HIV infection is characterized by continuous viral replication and depletion of
CD4 + T lymphocytes, leading to immunological changes and opportunistic
infections by pathogens. The use of antiretroviral therapy (ART) became
apparent regression of clinical manifestations, accompanied by improvement in
the patient's clinical condition. Fact which indicates an increase in life
expectancy. However, with the benefits of medication complications arise also
related to the toxicity of these drugs. HIV infection and HAART has been
strongly associated with the risk of cardiovascular disease and possible
prothrombotic events. Knowing the participation of platelets as effector cells in
the immune response, inflammatory and clotting and that the purinergic system
comprising ectoenzymes as E-NTPDase, and 5'-nucleotidase and E-ADA
besides their role in regulating levels of adenine nucleotides and nucleoside
have important actions in tromboregulation hemostasis and the aim of this study
was to determine the profile of platelet aggregation activity ectoenzymes in
platelets and metabolic changes in HIV-infected individuals. The study included
20 control subjects (tested negative for HIV), 10 HIV individuals (seropositive for
HIV without antiretroviral treatment) and 50 HIV-HAART (seropositive for HIV
and treated with HAART). The results show that there was no change in the
activity of the E-NTPDase for ATP hydrolysis, but there was a decrease in the
hydrolysis of ADP in HIV HAART group. The activity of E-5'-nucleotidase is not
changed to AMP hydrolysis in HIV HAART. However, the activity of E-ADA
groups is decreased in HIV and HIV HAART, which may promote an increase in
extracellular levels of adenosine. Also found increased platelet aggregation and
increased LDL cholesterol and triglyceride levels in HIV HAART group, these
data suggest increased risk of cardiovascular disease. Thus, the results
reported here suggest that antiretroviral treatment has a significant change,
both in the activity of the purinergic system ectoenzymes involved in
thromboregulation, as the profile of platelet aggregation in these patients and
metabolic parameters, data that can support risk hypotheses cardiovascular this
group of patients and a possible cardioprotective effect of extracellular adenosine during antiretroviral treatment. | eng |

dc.description.resumo | A infecção pelo HIV é caracterizada por uma contínua replicação viral e
depleção dos linfócitos T CD4+, acarretando alterações imunológicas e
infecções por patógenos oportunistas. A utilização da terapia antirretroviral
(TARV) tornou evidente a regressão das manifestações clínicas, acompanhada
por melhora nas condições de saúde do paciente. Fato que indica um aumento
na expectativa de vida. Entretanto, junto aos benefícios da medicação, surgem
também complicações relacionadas com a toxicidade destas drogas. A
infecção pelo HIV e o uso da TARV tem sido fortemente associados com o
risco de doença cardiovascular e possíveis eventos pró-trombóticos. Sabendo
da participação de plaquetas como células efetoras na resposta imune, na
resposta inflamatória e na coagulação sanguínea e ainda, que o sistema
purinérgico é constituído por ectoenzimas (E-NTPDase, E-5’-nucleotidase e EADA)
que possuem ações importantes na tromborregulação e na hemostasia o
objetivo deste estudo foi determinar o perfil de agregação plaquetária, a
atividade de ectoenzimas em plaquetas e possíveis alterações metabólicas de
indivíduos infectados pelo HIV. Foram incluídos no estudo 20 indivíduos
controle (sorologia negativa para HIV), 10 indivíduos HIV (sorologia reagente
para HIV e sem tratamento antirretroviral) e 50 indivíduos HIV TARV (com
sorologia reagente para HIV e em tratamento com TARV). Os resultados
mostram que não houve alteração da atividade da E-NTPDase para hidrólise
do ATP, mas houve uma diminuição de hidrólise do ADP no grupo HIV TARV.
A atividade da E-5’-nucleotidase não está alterada para hidrólise do AMP nos
pacientes HIV TARV. Contudo, a atividade da E-ADA está diminuída nos
grupos HIV e HIV TARV, fato que pode promover um aumento nos níveis
extracelulares de adenosina. Ainda foi encontrado aumento da agregação
plaquetária e aumento do colesterol LDL e triglicerídeos no grupo HIV TARV,
dados estes sugerem risco de doença cardiovascular aumentado. Com isso,
os resultados aqui relatados sugerem que o tratamento antirretroviral exerce
uma importante alteração, tanto na atividade das ectoenzimas do sistema
purinérgico envolvidas no processo tromborregulatório, quanto no perfil de
agregação plaquetária destes pacientes e nos parâmetros metabólicos, dados
que podem suportar hipóteses de risco cardiovascular neste grupo de
pacientes e um possível efeito cardioprotetor da adenosina extracelular durante
o tratamento antirretroviral. | por |
