dc.description.abstract | Mental illnesses are devastating illnesses that affect millions of people around the world. Depression and anxiety have significant financial and emotional costs for patients, families and society. Among the possible causes of these disorders are psychosocial stressors. Thus, exposure to acute restraint stress (ARS) represents an animal model for the induction of depressive and anxious-like behaviors, thus allowing the study of the pathophysiology of these disorders. In addition to the poor understanding of the neurobiology of these disorders, treatment is also limited, as one third of patients do not respond to treatment with antidepressants. In addition, these drugs have several side effects and can take weeks to take effect. Therefore, the discovery and innovation of new treatments/therapies are very important and necessary. Intermittent Hypoxia (IH) is a non-pharmacological alternative treatment that uses breathing with low oxygen concentration interspersed with normoxia periods. HI improves the body's general resistance, being used for pre-acclimatization to altitudes, in sports and in the treatment of various diseases, including mental disorders. Thus, the aim of this study was to investigate the anxiolytic and antidepressant potential, in addition to effects on serum corticosterone levels, oxidative parameters and inflammation, of an acute session (1 hour, 13% O2) of IH and its association with fluoxetine (FLX), in C57BL/6 mice submitted to the ARS model for 6 hours. The open field test (OFT), elevated plus maze test (EPMT), tail suspension test (TST) and forced swimming test (FST) were used to assess anxiety/depressive-like behaviors, which revealed anxiety-like behavior. Regarding oxidative stress, there was a decrease in hydrogen peroxide (H2O2) levels with the associated treatments compared to the control group and greater catalase (CAT) activity in the FLX treatment compared to IH, in the hippocampal tissue. Interleukin IL-17, IL-6 and IFN-γ levels increased after the ARS protocol. All treatments reversed the levels of IFN-γ, IL-17, IL-2 and TNF-α. IL-6 levels were altered by IH+FLX treatment. The results presented in this work demonstrate that ARS was insufficient to induce depressive-like behavior and that only 1 hour of IH and/or its association with fluoxetine did not reverse anxiety-like behavior, but induced changes in oxidative parameters and had an anti-inflammatory effect. | eng |

dc.description.resumo | Doenças mentais são doenças devastadoras que afetam milhões de pessoas no mundo. A depressão e a ansiedade apresentam significativos custos financeiros e emocionais para os pacientes, familiares e sociedade. Dentre as possíveis causas desses transtornos, encontram-se os agentes estressores psicossociais. Assim, a exposição ao estresse de contenção agudo representa um modelo animal para a indução dos comportamentos do tipo-depressivo e ansioso, permitindo assim estudar a fisiopatologia destes transtornos. Além do pobre entendimento da neurobiologia destes transtornos, o tratamento também é limitado, pois um terço dos pacientes não responde ao tratamento com antidepressivos. Além disso, esses medicamentos possuem diversos efeitos colaterais e podem demorar semanas para surtirem efeitos. Logo, a descoberta e inovação de novos tratamentos/terapias são muito importantes e necessárias. A Hipóxia Intermitente (HI) consiste em um tratamento alternativo não farmacológico que utiliza a respiração com baixa concentração de oxigênio intercalada com períodos normóxia. A HI melhora a resistência geral do organismo, sendo utilizada para pré-aclimatação a altitudes, no esporte e tratamento de várias doenças, dentre elas as desordens mentais. Assim, o objetivo deste estudo foi investigar o potencial ansiolítico e antidepressivo, além de efeitos sobre níveis séricos de corticosterona, parâmetros oxidativos e inflamação, de uma sessão aguda (1 hora, 13% O2) de HI e sua associação com fluoxetina (FLX), em camundongos C57BL/6 submetidos ao modelo de estresse de contenção agudo (ECA) por 6 horas. Para a avaliação dos comportamentos do tipo-ansioso/depressivo foram utilizados o teste de campo aberto (TCA), teste do labirinto em cruz elevado (TLCE), teste de suspensão da cauda (TST) e teste de nado forçado (TNF), que revelaram comportamento do tipo-ansioso. Em relação ao estresse oxidativo, houve diminuição dos níveis de peróxido de hidrogênio (H2O2) com os tratamentos associados em relação ao grupo controle e maior atividade da catalase (CAT) no tratamento FLX em relação a HI, no tecido hipocampal. Os níveis de interleucina IL-17, IL-6 e IFN-γ aumentaram após o protocolo de ECA. Todos os tratamentos reverteram os níveis de IFN-γ, IL17, IL-2 e TNF-α. Os níveis de IL-6 foram alterados pelo tratamento com HI+FLX. Os resultados apresentados neste trabalho demonstram que o ECA foi insuficiente para induzir comportamento do tipo-depressivo e que apenas 1 hora de HI e/ou sua associação com fluoxetina não reverteu o comportamento do tipo ansioso, mas induziu alterações nos parâmetros oxidativos e apresentou efeito anti-inflamatório. | por |
