dc.description.abstract | Concussion can be defined as a change in brain function caused by an external force. In recent years, it has received greater attention from the scientific community for its subtle effects, which can add up and worsen the secondary damage of the injury. It’s very common in the context of sports initiation in schools, recurrent concussions (RC) lack studies that evaluate their long-term effects on the central nervous system (CNS). Interest is growing about the effects of CR in adolescence can influence neural architecture and metabolism in adulthood, in different situations and pathologies. Thus, to investigate this knowledge gap, the present work submitted 35-day-old rats to a protocol of 7 CR (one per day). The analysis of the behavioral tests applied 30 days after the last CR revealed that there was a decrease in short- and long-term memory in the object recognition test and, in addition, there was no change in motor and anxious behavior. In addition, the CR protocol induced 34 days after the last insult the inflammatory/oxidative stress characterized by the increase of glial fibrillary acidic protein (GFAP), interleukin-1β (IL-1β), 4-hydroxynonenal (4-HNE), immunoreactivity of protein carbonyl and oxidation of 2',7'-dichlorofluorescein diacetate (DCFH) and lower total antioxidant capacity (TAC). In addition, through statistical analysis it was also revealed that CR alter the activity and expression of the enzyme Na+, K+-ATPase, specifically, the inhibited Na+,K+-ATPase activity (α2/3 isoform) and decreased immunodetection of the alpha subunit of this enzyme. With these results, it is plausible to propose that cognitive impairment after CR is caused by the inability of neurons to survive because they cannot maintain ionic gradients in selected targets for inflammatory/oxidative damage, such as Na,K-ATPase enzyme activity. | eng |

dc.description.resumo | A concussão pode ser definida como uma alteração na função cerebral ocasionada por uma força externa. Nos últimos anos, tem recebido maior a atenção da comunidade científica por seus efeitos sutis, que podem se somar e agravar o dano secundário da lesão. Muito comum no contexto de iniciação esportiva nas escolas, as concussões recorrentes (CR) carecem de estudos que avaliem seus efeitos a longo prazo no sistema nervoso central (SNC). É crescente o interesse acerca dos efeitos de CR na fase da adolescência pode influenciar na arquitetura e no metabolismo neural na vida adulta, em diferentes situações e patologias. Dessa forma, para investigar essa lacuna do conhecimento o presente trabalho submeteu ratos com 35 dias de idade a um protocolo de 7 CR (uma por dia). As análises dos testes comportamentais aplicados 30 dias após a última CR revelaram que houve uma diminuição na memória de curto e longo prazo no teste de reconhecimento de objetos e, além disso, não houve alteração no comportamento motor e ansioso. Além disso, o protocolo de CR induziu 34 dias após o último insulto o estresse inflamatório/oxidativo caracterizado pelo aumento da proteína glial fibrilar ácida (GFAP), interleucina-1β (IL-1β), 4-hidroxinonenal (4-HNE), imunorreatividade da proteína carbonil e oxidação do diacetato de 2',7'-diclorofluoresceína (DCFH) e menor capacidade antioxidante total (TAC). Além disso, através da análise estatística também foi revelado que as CR alteram a atividade e a expressão da enzima Na+, K+-ATPase, especificamente, a atividade Na+,K+-ATPase inibida (isoforma α2/3) e diminuição da imunodetecção da subunidade alfa desta enzima. Com esses resultados, é plausível propor, que o comprometimento cognitivo após CR é causado pela incapacidade dos neurônios sobreviverem por não conseguirem manter os gradientes iônicos em alvos selecionados para danos inflamatórios/oxidativos, como atividade da enzima Na,K-ATPase. | por |
