dc.description.abstract | Baculoviruses are bioinsecticides widely used in Brazil and in the world and can
replace, assist or reduce the consumption of agrochemicals in agricultural crops. They
are part of the Baculoviridae family, a family of viruses that contain a double-stranded,
supercoiled, circular DNA genome. The genome size varies between 80-180 kb and
encodes 90-180 genes, of which only 38 are present in all baculoviruses sequenced to
date. They are insect-specific pathogens and are divided into four genera,
Alphabaculovirus and Betabaculovirus contain members that are infective for
lepidopteran larvae, Gammabaculovirus contain members that are infective for
Hymenopteran larvae, and Deltabaculovirus which contain members that are infective
for dipteran larvae. In addition, as a distinct characteristic, they present two viral
phenotypes during a complete cycle of infection, one responsible for the primary oral
infection and which is protected within the body from occlusion, active principle of
baculovirus-based bioinsecticides and another phenotype responsible for the spread of
secondary infection along the body of the insect. In the present work, the genome of a
new baculovirus isolated from the rice and wheat pest of the Mythimna sequax species
that causes an economic impact on these crops was characterized. Scanning electron
microscopy confirmed the characteristic polyhedral occlusion body of
Alphabaculoviruses, hence the virus was named Mythimna sequax
nucleopolyhedrovirus (MyseNPV). The virus genome showed 148,403 bp, G + C
content of 40.3% and 169 genes were noted, 13 of which are exclusive to this
baculovirus. It is a member of the genus Alphabaculovirus and is closely related to
seven other baculoviruses that infect insects of the family Noctuidae, including the
viruses that infect insects of the genus Mamestra and the species Helicoverpa armigera.
Although there are five recognized species among those closely related, only three were
true species, MyseNPV being potentially one of them. The other viruses were just viral
isolates. This was reinforced by the similarity of the chitinase and cathepsin gene locus
between the true species. A selection analysis was performed for each gene shared by
all MyseNPV-related genomes using an individual dN/dS analysis. A total of 221
different genes were found with only 152 shared by the eight genomes. Only three
genes underwent positive selection in this group, hoar, bro-d and MyseNPV-127. The
fp25k, p28, cathepsin, vlf-1 are those that undergo the greatest purification selection of
those 152 genes. That said, the increase in the number of fully sequenced baculoviruses
is beneficial for a better understanding of baculoviruses and their evolution, potentially
helping the use of baculoviruses both as a biopesticide and expression vectors. | eng |

dc.description.resumo | Os baculovírus são bioinseticidas amplamente utilizados no Brasil e no mundo e podem
substituir, auxiliar ou reduzir o consumo os agroquímicos comuns em lavouras
agrícolas. Fazem parte da família Baculoviridae, uma família de vírus que contém
genoma de DNA circular, superenovelado de fita dupla. O tamanho do genoma varia
entre 80-180 kb e codifica para 90-180 genes, destes apenas 38 estão presentes em todos
os baculovírus sequenciados até a data. Eles são patógenos específicos para insetos e
são divididos em quatro gêneros, Alphabaculovirus e Betabaculovirus contém membros
que são infectivos para larvas de lepidópteros, Gammabaculovirus contém membros
que são infectivos para larvas de himenópteros e Deltabaculovirus que contém
membros infectivos para larvas de dípteros. Além disso, como característica distinta,
apresentam dois fenótipos virais durante um ciclo completo de infecção, um responsável
pela infecção oral primária e que está protegido dentro do corpo de oclusão, princípio
ativo de bioinseticidas a base de baculovírus e outro fenótipo responsável pelo
espalhamento da infecção secundária ao longo do corpo do inseto. No presente trabalho,
foi caracterizado o genoma de um novo baculovírus isolado da praga de arroz e trigo da
espécie Mythimna sequax que causa impacto econômico nessas culturas agrícolas.
Microscopia eletrônica de varredura confirmou o corpo de oclusão poliédrico
característico dos Alphabaculovirus, por isso o vírus foi denominado Mythimna sequax
nucleopolyhedrovirus (MyseNPV). O genoma do vírus apresentou 148.403 pb,
conteúdo G + C de 40,3% e foram anotados 169 genes, sendo 13 exclusivos deste
baculovírus. Trata-se de um membro do gênero Alphabaculovirus e está proximamente
relacionado a outros sete baculovírus que infectam insetos da família Noctuidae,
incluindo os vírus que infectam insetos do gênero Mamestra e da espécie Helicoverpa
armigera. Apesar de haver cinco espécies reconhecidas dentre os proximamente
relacionados, apenas três eram espécies verdadeiras, sendo MyseNPV potencialmente
uma delas. Os outros vírus eram apenas isolados virais. Isso foi reforçado pela
similaridade do lócus dos genes da chitinase e cathepsin entre as espécies verdadeiras.
Realizou-se uma análise de seleção para cada gene compartilhado por todos os genomas
relacionados ao MyseNPV por meio de uma análise de dN/dS individual. Um total de
221 genes diferentes foram encontrados com apenas 152 compartilhados pelos oito
genomas. Apenas três genes sofriam seleção positiva neste grupo, hoar, bro-d e
MyseNPV-127. O fp25k, p28, catepsina, vlf-1 são aqueles que sofrem maior seleção de
purificação daqueles 152 genes. Dito isso, o aumento no número de baculovírus
completamente sequenciados é benéfico para o melhor entendimento dos baculovírus e
a sua evolução, podendo ajudar potencialmente o uso de baculovírus tanto como
biopesticida, como vetores de expressão. | por |
