Caracterização de Corynebacterium sp. isolado do útero de éguas do sul do Brasil
Abstract
A população de equinos no Rio Grande do Sul é expressiva, totalizando aproximadamente 500 mil cabeças em 2021. Entre as atividades exercidas nessa espécie está a reprodução. Por isso, os problemas reprodutivos podem representar grandes perdas econômicas e genéticas. Uma das principais responsáveis por essas desordens é a endometrite, que pode ser fisiológica, mas que pode evoluir para um processo patológico e, por consequência, reduzir a fertilidade das éguas. Muitos fatores podem estar associados à endometrite, principalmente, a infecção por microrganismos como Streptococcus equi subsp. zooepidemicus e Escherichia coli. Em 2013, uma nova espécie bacteriana foi isolada de éguas com problemas reprodutivos na Europa, e foi denominada Corynebacterium uterequi. Em 2022, esse microrganismo foi isolado de éguas da África do Sul que também apresentavam problemas de fertilidade. Uma vez que ainda se desconhece o real envolvimento dessa bactéria na ocorrência de lesões uterinas e infertilidade em éguas, a sua distribuição e características devem ser investigadas. Portanto, o objetivo desse estudo foi caracterizar Corynebacterium sp. isolado de amostras uterinas de éguas com problemas reprodutivos em um haras do Rio Grande do Sul, e descrever os achados citológicos e histopatológicos de éguas positivas para esse microrganismo. Para isso, 30 amostras uterinas coletadas de éguas de um haras de São Borja (RS), com histórico de problemas reprodutivos, foram submetidas à análise laboratorial (cultura, citologia e histopatologia). Para a cultura bacteriana, as amostras de suabe foram inoculadas em ágar enriquecido com sangue de carneiro a 5% e ágar MacConkey. Após 48h de incubação em aero e microaerobiose, Corynebacterium sp. foi isolado de doze amostras, associado ou não a outros microrganismos. Na análise morfológica e bioquímica, o isolado apresentou características altamente similares ao de C. uterequi obtido de éguas da Europa e da África do Sul. A análise molecular das regiões 16S RNA e rpoB indicou que o isolado possui alta identidade com C. uterequi depositados no Genbank (99,38% e 100%, respectivamente). Na avaliação citológica do útero de éguas positivas para Corynebacterium sp. foi observada celularidade indicativa de inflamação em oito amostras (8/12). Na biopsia de útero, em seis amostras (6/12) foram observadas alterações compatíveis com fibrose glandular e periglandular, em um caso havia formação de ninho periglandular e ectasia glandular e, em outro, foram observadas lesões características de endometrite linfoplasmocítica. Esses resultados não permitem estabelecer se C. uterequi isolado de éguas do Rio Grande do Sul pertence à microbiota ou se é um agente causador de lesões, já que foi detectado tanto em animais com alterações uterinas quanto sem alterações histológicas. No entanto, este trabalho contribuiu para o conhecimento da epidemiologia, clínica, patologia e diagnóstico desta nova espécie, além de demonstrar sua circulação em éguas do sul do Brasil.
