dc.description.abstract | Gestational diabetes mellitus (GDM) is a glucose intolerance beginning during pregnancy. It is a common disorder and its incidence is increasing among pregnant women. Hyperglycemia derived from GDM is associated with the formation of reactive oxygen species (ROS) in excess, which are capable of increasing oxidative stress and damage the health of both mother and fetus. The δ-aminolevulinate dehydratase enzyme (δ-ALA-D) is sensitive to oxidative conditions such as hyperglycemia. The aim of this study was to evaluate oxidative stress biomarkers in pregnant women with GDM and their relation to the activity of δ-ALA-D in order to analyze the oxidative profile of these pregnant women in the course of this disease. The evaluation was performed in 48 pregnant women with GDM and 30 healthy pregnant women (controls), both groups were in the third trimester. The activity of δ-ALA-D was measured in whole blood sample. Lipid peroxidation was evaluated by measuring thiobarbituric reactive substances (TBARS) in plasma and erythrocytes. Already for the analysis of antioxidant status were quantified levels of vitamin C (VIT C) in plasma, catalase (CAT) enzyme in erythrocytes, and the protein thiol groups (P-SH) in plasma and non-protein (NP-SH) in erythrocytes. The results showed a reduction of the δ-ALA-D activity, as well as an increase in its reactivation index in pregnant women with GDM compared to controls. Furthermore, there was an increase in TBARS levels in the GDM group (p<0.05). Antioxidants evaluated in pregnant women with GDM exhibited reduced levels compared to controls (p<0.05). In addition, there were significant correlations between δ-ALA-D and levels of NP-SH, reactivation index and erythrocyte TBARS; the latter was also correlated with fasting glucose levels in women with GDM. Therefore, there was an increase of oxidative stress in women with GDM, unlike what happens in a normoglycemic pregnancy. In parallel with increased oxidative stress was observed a reduction of the δ-ALA-D activity, which was shown to be sensitive to the hyperglycemic environment that emerged during pregnancy. Thus, the use of δ-ALA-D together with other markers of oxidative stress may be important to assess metabolic processes that are debilitated, as in GDM. | eng |

dc.description.resumo | O Diabetes mellitus gestacional (DMG) é uma intolerância à glicose com início durante a gestação. É um distúrbio frequente e sua incidência está crescendo entre as gestantes. A hiperglicemia resultante do DMG está associada à formação de espécies reativas de oxigênio (EROs) em excesso, as quais são capazes de aumentar o estresse oxidativo e prejudicar a saúde tanto materna quanto fetal. A enzima δ-Aminolevulinato desidratase (δ-ALA-D) é sensível a situações oxidativas, como a hiperglicemia. O objetivo deste trabalho foi avaliar biomarcadores de estresse oxidativo em gestantes com DMG e sua relação com a atividade da δ-ALA-D, a fim de analisar o perfil oxidativo dessas gestantes no transcurso desta doença. A avaliação foi realizada em 48 gestantes com DMG e 30 gestantes saudáveis (controles), ambos os grupos estavam no terceiro trimestre gestacional. A atividade da δ-ALA-D foi mensurada em amostra de sangue total. A peroxidação lipídica foi avaliada através da quantificação das substâncias reativas ao ácido tiobarbitúrico (TBARS) no plasma e em eritrócitos. Já para a análise do perfil antioxidante foram quantificados os níveis de vitamina C (VIT C) no plasma, da enzima catalase (CAT) em eritrócitos, e dos grupos tióis protéicos (P-SH) em plasma e não-protéicos (NP-SH) em eritrócitos. Os resultados demonstraram uma redução da atividade da δ-ALA-D, assim como um aumento de seu índice de reativação nas gestantes com DMG quando comparadas as controles (p<0,05). Além disso, houve aumento dos níveis de TBARS no grupo DMG (p<0,05). Os antioxidantes avaliados nas gestantes com DMG apresentaram níveis reduzidos quando comparados as controles (p<0,05). Além disso, houve correlações significativas entre δ-ALA-D e os níveis de NP-SH, índice de reativação e TBARS de eritrócitos; este último também foi correlacionado com os níveis de glicose de jejum em mulheres com diabetes gestacional. Portanto, houve um aumento do estresse oxidativo em mulheres com DMG, diferente do estresse de uma gestação normoglicêmica. Em paralelo ao aumento do estresse oxidativo foi observada uma redução da atividade da δ-ALA-D, a qual demonstrou ser sensível ao ambiente hiperglicêmico que surgiu durante a gestação. Assim, a utilização de δ-ALA-D em conjunto com outros marcadores de estresse oxidativo pode ser importante para avaliar processos metabólicos que estão debilitados, como no DMG. | por |
