dc.description.abstract | The present dissertation aims to contribute to correct knowledge of biological activities of Nectandra
grandiflora Nees & Mart. ex Nees essential oil (EO) and its major compound, (+)-dehydrofukinone
(DHF). So, the potential this extract was verified to act as biostimulant in native woody species explants
cultivated in vitro. Additionally, the antifungal potential of both N. grandiflora OE and its major constituent
against wood rot fungi was determined, as well as its action on ergosterol content in the plasma
membrane of phytopathogens. The EO was obtained from N. grandiflora leaves by hydrodistillation with
modified Clevenger apparatus. Then, extraction yield was determined, and chemical composition was
analyzed by gas chromatography coupled to mass spectrometry (GC-MS). The major constituent of OE
was isolated by 4 chromatographic columns, carried out in sequence. For tests with the explants,
Eugenia involucrata De Candolle nodal segments were inoculated in ½ MS medium, supplemented with
N. grandiflora EO, previously diluted in ethanol, at .0.00, 0.05, 0.075, 0.1, 0.125, 0.15, 0.175 e 0.2 μL
mL-1. The results showed that, when exposed to EO at 0.075 μL mL-1, the explants emitted a greater
number of leaves when compared to the controls. For determination of N. grandiflora OE and its major
constituent antifungal potential, the extractives previously diluted in ethanol were added to PDA medium.
The antifungal potential was verified against white-rot fungi Ganoderma applanatum (Pers.) Pat. and
Trametes versicolor (L.: Fr) Pilát, as well as brown-rot fungi Lentinus lepideus (Fr.) Fr. and Gloeophyllum
trabeum (Pers.: Fr) Murrill. The EO was evaluated at 0.0, 0.5, 1.0 and 2.0 μL mL-1, whereas DHF was
evaluated at the corresponding concentration to that found in 0.5 μL mL-1 EO (0.0971 μL mL-1).
Regarding the results of mycelium growth inhibition, N. grandiflora EO completely inhibited growth of
white-rot fungi T. versicolor, at 1.0 μL mL-1 EO, presenting the same behavior as positive control. The
other fungal species did not have their growth affected by the extractives at the tested concentrations.
Additionally, the results for the EO treatments did not differ from those obtained with the isolated
substance, evidencing that the DHF is one of the substances responsible for the antifungal activity
detected. Regarding the influence of the extractives on ergosterol content in plasma membrane of
phytopathogens, the results indicated that both extractives evaluated caused a reduction of this sterol
in T. versicolor and G. applanatum, indicating that their action target was the plasma membrane of whiterot
fungi. Thus, the potential of N. grandiflora EO to act as possible biostimulant in the development of
in vitro nodal segments of E. involucrata is highlighted, as well as to inhibit the growth of white-rot fungus

dc.description.resumo | A presente dissertação visa contribuir com o conhecimento acerca das atividades biológicas do óleo
essencial (OE) de Nectandra grandiflora Nees & Mart. ex Nees e seu constituinte majoritário, a
(+)-deidrofuquinona (DHF). Para isto, verificou-se o potencial deste extrativo para atuar como
bioestimulante em explantes de uma espécie lenhosa nativa cultivada in vitro. Adicionalmente,
determinou-se o potencial antifúngico tanto do OE de N. grandiflora quanto do seu constituinte
majoritário frente a fungos apodrecedores da madeira, além de verificar sua ação sobre o conteúdo de
ergosterol na membrana plasmática dos fitopatógenos. O OE foi obtido a partir das folhas de N.
grandiflora por meio de hidrodestilação, com o auxílio do aparelho tipo Clevenger modificado. Em
seguida, determinou-se o rendimento de extração e realizou-se a análise da composição química por
cromatografia gasosa acoplada a espectrometria de massas (CG-EM). O constituinte majoritário do OE
foi isolado por meio de 4 colunas cromatográficas, realizadas em sequência. Para os testes com os
explantes, segmentos nodais de Eugenia involucrata De Candolle foram inoculados em meio nutritivo
½ MS, suplementado com o OE de N. grandiflora, previamente diluído em etanol, nas concentrações
de 0,00, 0,05, 0,075, 0,10, 0,125, 0,150, 0,175 e 0,200 μL mL-1. Os resultados evidenciaram que, na
concentração de 0,075 μL mL-1, os explantes emitiram um maior número de folhas quando comparados
aos controles. Para a determinação do potencial antifúngico do OE de N. grandiflora e de seu
constituinte majoritário, os extrativos, previamente diluídos em etanol, foram adicionados ao meio
nutritivo BDA. O potencial antifúngico foi verificado frente aos fungos causadores de podridão-branca
Ganoderma applanatum (Pers.) Pat. e Trametes versicolor (L.: Fr) Pilát, bem como frente aos fungos
causadores de podridão-parda Lentinus lepideus (Fr.) Fr. e Gloeophyllum trabeum (Pers.: Fr) Murrill. O
OE foi avaliado nas concentrações de 0,0, 0,5, 1,0 e 2,0 μL mL-1, enquanto que a DHF foi avaliada na
concentração correspondente à encontrada em 0,5 μL mL-1 de OE (0,0971 μL mL-1). Quanto aos
resultados de porcentagem de inibição do crescimento micelial, destaca-se que o OE de N. grandiflora
inibiu totalmente o crescimento do fungo de podridão-branca T. versicolor a 1,0 μL mL-1, apresentando
o mesmo comportamento que o controle positivo. As demais espécies fúngicas não tiveram seu
crescimento afetado pelos extrativos nas concentrações avaliadas. Adicionalmente, os resultados
encontrados nos tratamentos com OE não diferiram daqueles realizados com a substância isolada,
evidenciando que a DHF é uma das substâncias responsáveis pela atividade antifúngica detectada. No
que tange à influência dos extrativos sobre o conteúdo de ergosterol na membrana plasmática dos
fitopatógenos, os resultados indicaram que ambos os extrativos avaliados causaram uma redução
deste esterol para os fungos T. versicolor e G. applanatum, indicando ter como alvo de ação a
membrana plasmática dos fungos causadores de podridão-branca. Com base no exposto, destaca-se
o potencial do OE de N. grandiflora para atuar como um possível bioestimulante no desenvolvimento
de segmentos nodais cultivados in vitro de E. involucrata, bem como para inibir o crescimento do fungo
apodrecedor da madeira T. versicolor. | por |
