dc.description.abstract | The entomopathogenic fungus Metarhizium anisopliae is used for the biological control of agricultural pests due to the secretion of a complex of hydrolytic enzymes such as chitinase and β-1,3-glucanase. In recent years, much attention has been paid to the use of these enzymes as an active ingredient in bioproducts. Thus, this study was focused on the production of hydrolytic enzymes by solid state fermentation (SSF) and its drying by spray drying. Different substrates (sugarcane bagasse, malt bagasse and white rice) supplemented with corn steep liquor, soybean meal and chitin were evaluated for enzymatic production by SSF with the fungus Metarhizium anisopliae IBCB 348. Sugarcane bagasse was the best substrate for enzymatic production, with activities of 12.07, 11.54, 20.87 and 22.30 U/g for chitinase, β-1,3-glucanase, endocellulase and exocellulase, respectively. The best conditions of SSF for enzymatic production were moisture content of 40%, inoculum concentration of 10% (v/w), temperature of 28 °C and 8 days of fermentation. Evaluating the spray drying process of the enzymatic broth obtained by SSF, the combination of adjuvants maltodextrin (2.5% w/v), arabic gum (2.5% w/v) and soluble starch (5.0% w/v) presented the best results of residual activities of β-1,3-glucanase (88.36%) and chitinase (69.82%), and a powder recovery yield of 45.49%. The operational parameters of the spray drying process were optimized using a Central Composite Rotational Design (CCRD), with the optimum conditions being: drying air inlet temperature of 120 °C, drying air flow rate of 1.1 m³/min, feed flow rate of 5.8 mL/min and atomizer air pressure of 0.4 MPa. The powder produced in the optimized conditions showed 65.6% efficiency in the control of Ceratitis capitata. These results demonstrated the potential of SSF for the production of hydrolytic enzymes and the possibility of its drying by spray drying. | eng |

dc.description.resumo | O fungo entomopatogênico Metarhizium anisopliae é utilizado no controle biológico de pragas agrícolas devido à produção de um complexo de enzimas hidrolíticas como quitinase e β-1,3-glucanase. Nos últimos anos, muita atenção tem sido dada ao uso destas enzimas como ingrediente ativo em bioprodutos. Assim, o presente estudo teve como principal objetivo a produção de enzimas hidrolíticas pelo processo de fermentação em estado sólido (FES) e a sua secagem por spray drying. Diferentes substratos (bagaço de cana-de-açúcar, bagaço de malte e arroz branco) suplementados com água de maceração de milho, farelo de soja e quitina foram avaliados para a producão enzimática por FES com o fungo Metarhizium anisopliae IBCB 348. O bagaço de cana foi o melhor substrato para a produção enzimática, com máximas atividades de 12,07, 11,54, 20,87 e 22,30 U/g para quitinase, β-1,3-glucanase, endocelulase e exocelulase, respectivamente. As melhores condições do processo de FES para produção enzimática foram teor de umidade de 40%, concentração de inóculo de 10% (v/m), temperatura de 28 °C e 8 dias de fermentação. Ao avaliar o processo de spray drying do caldo enzimático obtido por FES, a combinação dos adjuvantes maltodextrina (2,5% m/v), goma arábica (2,5% m/v) e amido solúvel (5,0 % m/v) apresentou os melhores resultados de atividades residuais de β-1,3-glucanase (88,36%) e quitinase (69,82%), e um rendimento de 45,49% na recuperação de produto final. Os parâmetros operacionais do processo de spray drying foram otimizados utilizando um Delineamento Composto Central Rotacional (DCCR), sendo as condições ótimas: temperatura de entrada do ar de secagem de 120 °C, vazão do ar de secagem de 1,1 m³/min, vazão de alimentação de 5,8 mL/min e pressão de ar do atomizador de 0,4 MPa. O pó produzido nas condições ótimas apresentou 65,6% de eficiência no controle de Ceratitis capitata. Estes resultados demonstraram o potencial da produção de enzimas hidrolíticas por FES e a possibilidade de sua secagem utilizando o processo de spray drying. | por |
