dc.description.abstract | Proliferation, functional differentiation and morphological transformation of granulosa cells (GCs) are events that occur for ovarian follicular development, achieved, atresia and ovulation. There is evidence to show that GC apoptosis is the main mechanism of ovarian follicular atresia. In previous studies, it was shown that the inhibitory effect of FGF18 on steroidogenesis is similar to what occurs in atretic follicles compared to dominant follicles. Assuming that FGF18 may be involved in the process of steroidogenesis, directly affecting the expression of effectors of the Hippo signal pathway in GCs and regulating the expression of key genes that control this event, we relate the function of the Hippo signal pathway with FGF18 and a adjusting for folliculogenesis in bovine GCs and analyzing different targets of YAP1-TEAD interaction to demonstrate, in vitro, the importance of the Hippo pathway in the process of follicular divergence. In order to establish this relationship, experiments were carried out with granulosa cells from ovarian follicles of female cows from slaughterhouses. The experiments were carried out with different concentrations of FGF18 (0, 1, 10 and 100 ng/mL) controlled on the fifth day of cultivation. Granulosa cells were cultured for six days in 24-well plates at a density of 0.5x10 -6 cells per well in modified DMEN medium free of fetal bovine serum. First day of cultivation, characterized as day zero, the cells were maintained only with insulin (10 ng/mL), remaining until the first change of medium. On day two and day four of cultivation, 70% of the culture medium was changed and treatment with FSH (1ng/mL), insulin (10ng/mL) was added, and on the fifth day treatment with different concentrations was added of FGF18 (0, 1, 10 and 100 ng/ml). Observe a decrease in mRNA levels of CTGF and CYR61, specific expression genes when the YAP protein is found in the cell nucleus and the Hippo signal pathway is inactive, and a decrease in the expression of the BAX and BCL2, indicating the occurrence of cell apoptosis. STAR and CYP19 mRNA expression was not significantly different. Another important gene that it was possible to observe the reduction of mRNA expression is GFPT2, which indicates the reduction of hexosamine metabolism, an important amine for cells to be able to exercise their physiological functions and their metabolic effects. As the translocation of YAP is very dynamic, it is believed that the time of treatment of cells with FGF18 may have influenced the results obtained, as there are studies that indicate that with 12 hours of treatment there is an increase in CTGF, that is, how GCs were being treated for 24 hours it was already going into apoptosis and there was a decrease in the expression of CTGF, an external cell gene. | eng |

dc.description.resumo | Proliferação, diferenciação funcional e transformação morfológica de células da granulosa (CGs) são eventos significativos para o desenvolvimento folicular ovariano, maturação, atresia e ovulação. Há evidências que mostram que a apoptose das GCs é o principal mecanismo da atresia folicular ovariana. Em estudos anteriores, foi demonstrado que o efeito inibitório do FGF18 na esteroidogênese se assemelha ao que ocorre em folículos atrésicos em comparação com folículos dominantes. Assumindo que o FGF18 pode estar envolvido no processo de esteroidogênese, afetando diretamente a expressão de efetores da via de sinalização Hippo em GCs e regulando a expressão de genes-chave que controlam esse evento, relacionamos a função da via de sinalização Hippo com o FGF18 e a regulação da foliculogênese em GCs bovinos e analisamos diferentes alvos de interação YAP1-TEAD para demonstrar, in vitro, a importância da via Hippo no processo de divergência folicular. Com o intuito de estabelecer essa relação, realizou-se experimentos com células da granulosa de folículos ovarianos de fêmeas boninas oriundas de abatedouros. Os experimentos foram realizados com diferentes concentrações de FGF18 (0, 1, 10 e 100 ng/mL) administradas no quinto dia de cultivo. As células da granulosa foram cultivadas durante seis dias em placas de 24 poços com densidade de 0,5x10-6 células por poço em meio DMEN modificado livre de soro fetal bovino. Primeiro dia do cultivo, caracterizado como dia zero, as células foram mantidas apenas com insulina (10 ng/mL), permanecendo até a primeira troca de meio. No dia dois e no dia quatro do cultivo, foi realizada a troca de 70% do meio de cultivo e acrescentou tratamento com FSH (1ng/mL), insulina (10ng/mL) e no quinto dia foi adicionado o tratamento com as diferentes concentrações de FGF18 (0, 1, 10 e 100 ng/mL). Observou-se uma diminuição nos níveis de mRNA de CTGF e CYR61, genes de proliferação expressos quando a proteína YAP encontra-se no núcleo celular e a via de sinalização Hippo está inativa, e uma diminuição da expressão dos genes BAX e BCL2, indicando a ocorrência da apoptose das células. A expressão de mRNA de STAR e CYP19 não tiveram diferença significativa. Outro gene importante que foi possível observar a redução da expressão RNAm é o GFPT2, que indica a redução do metabolismo hexosamina, uma amina importante para que as células consigam exercer suas funções fisiológicas e seus efeitos metabólicos. Como a translocação de YAP é muito dinâmica, acredita-se o tempo de tratamento das células com FGF18 pode ter influenciado nos resultados obtidos, pois há estudos que indicam que com 12 horas de tratamento há um aumento de CTGF, ou seja, as CGs estavam sendo tratadas por 24 horas já estavam entrando em apoptose e houve uma diminuição na expressão de CTGF, um gene de proliferação celular. | por |
