dc.description.abstract | The epithelial-mesenchymal transition (TEM) is a fundamental cellular process based
on the conversion of epithelial cells into mesenchymal cells, which plays an essential
role in the dissemination of cancer through metastasis. The evolution of knowledge
in terms of the molecules and interactions that govern the complex TEM system has
required the use of computational approaches that provide non-intuitive results on the
complexity of its dynamics. Thus, this study aims to analyze recent biochemical information
published in the literature through logical computational modeling in order to
amplify the dynamic knowledge of the transition. The two proposed models are related
to TEM activating molecule TGF-β. The data were compared with theoretical and
experimental data for MCF10A human breast epithelial cell line presenting excellent
agreement. Collectively, the results demonstrated the functionality of the circuits during
the transition and, thus, microRNAs 129, 340 and 1199 may be considered factors
of phenotypic stability of the hybrid state. Additionally, the data revealed the phenotypic
probabilities in the coexistence region and demonstrated that the hybrid state is the
most probable in the tristable dynamics, agreeing with experimental data. Therefore,
the present study highlights new mechanisms related to the dynamics of TEM in response
to the TGF-β molecule in breast epithelial cells and such results help to improve
new therapeutic strategies for breast cancer. | eng |

dc.description.resumo | A transição epitélio-mesenquimal (TEM) é um processo celular fundamental baseado
na conversão de células epiteliais em mesenquimais, que desempenha um papel essencial
na disseminação do câncer através da metástase. A evolução do conhecimento
em termos das moléculas e interações que governam o complexo sistema da
TEM tem requerido a utilização de abordagens computacionais que proporcionam resultados
não intuitivos sobre a complexidade de sua dinâmica. Assim, este estudo
objetiva analisar recentes informações bioquímicas publicadas na literatura através da
modelagem computacional lógica de maneira a amplificar o conhecimento dinâmico da
transição. Os dois modelos propostos no presente estudo são relacionados à molécula
ativadora da TEM TGF-β. Seus resultados foram comparados com dados teóricos e
experimentais para a linhagem de células epiteliais da mama humana MCF10A apresentando
excelente concordância. Coletivamente, os resultados demostraram a funcionalidade
dos circuitos durante a transição e, dessa forma, os microRNAs 129, 340
e 1199 podem ser considerados fatores de estabilidade fenotípica do estado híbrido.
Adicionalmente, os dados revelaram as probabilidades fenotípicas na região de coexistência
e constatam que o estado híbrido é o mais provável na dinâmica tri-estável,
concordando com dados experimentais. Portanto, o presente estudo demonstra novos
mecanismos relacionados à dinâmica da TEM em resposta à molécula TGF-β em
células epiteliais da mama e tais resultados auxiliam no aprimoramento de novas estratégias
terapêuticas para o câncer de mama. | por |
