dc.description.abstract | Sports practice can induce injuries to muscle fibers, generating oxidative damage and
an inflammatory process. Therapeutic practices are used to reduce this damage, such
as platelet-rich plasma (PRP) and cold-water immersion (CWI). Thus, the present
study aims to investigate through biomarkers the influence of treatment with
therapeutic cold and/or PRP in experimental muscle injury models in animals. In the
first study, we present the therapeutic effects of PRP after muscle contusion, and we
observed that the contusion increased the levels of oxidative stress markers, such as
thiobarbituric acid reactive substances and oxidized dichlorofluorescein, both in
skeletal muscle tissue and in erythrocyte preparations, and PRP treatment reduced
these markers of oxidative damage. In addition, contusion decreased cell viability at
the injury site and PRP was effective in modulating this effect, as it was also able to
increase the activity of antioxidant enzymes (superoxide dismutase and catalase) in
the injured muscle, as well as the levels of the non-protein thiols group (-SH) in
erythrocytes. In a second study, we compared the combination of two treatments (PRP
and CWI) very commonly used in sports. The levels of thiobarbituric acid-reactive
substances and oxidized dichlorofluorescein were significantly increased in both
skeletal muscle tissue and erythrocytes, and the combination of PRP and CWI
minimized these parameters. In addition, PRP and CWI combined were more effective
than treatments alone in increasing catalase activity, reduced/oxidized glutathione
ratio, and non-protein thiol (-SH) group levels. Finally, in the third work, we compared
the two methods of experimental muscle injury used in the two previous works. The
contusion and strain injury increased the levels of oxidative stress markers, however,
the injuries showed significant differences from each other in the markers of oxidized
dichlorofluorescein, superoxide dismutase enzyme activity and N-acetyl-beta
glucosaminidase enzyme activity in skeletal muscle tissue. In addition, the lesions
showed differences in the behavioral analyzes in the bar walk test. In general, the
results obtained in this thesis support a new strategy for the treatment of muscle
injuries using the combination of PRP and CWI. In addition, our studies support the
understanding of the specificity of these musculoskeletal injuries, supporting treatment
strategies that can effectively rehabilitate these sports injuries, minimizing
unnecessary costs and accelerating the return of athletes to their practice. | eng |

dc.description.resumo | A prática esportiva pode induzir lesões nas fibras musculares, gerando um dano
oxidativo e processo inflamatório. Práticas terapêuticas são utilizadas com o intuito de
reduzir estes danos, como o plasma rico em plaquetas (PRP) e a imersão em água
fria (CWI). Assim, o presente estudo tem como objetivo investigar através de
biomarcadores a influência do tratamento com o frio terapêutico e/ou PRP em modelos
de lesões musculares experimentais em animais. No primeiro trabalho apresentamos
os efeitos terapêuticos do PRP após a contusão muscular, e observamos que a
contusão aumentou os níveis de marcadores de estresse oxidativo, como substâncias
reativas ao ácido tiobarbitúrico e diclorofluoresceína oxidada, tanto no tecido muscular
esquelético quanto nas preparações de eritrócitos, e o tratamento com PRP reduziu
esses marcadores de dano oxidativo. Além disso, a contusão diminuiu a viabilidade
celular no local da lesão e o PRP foi eficaz em modular esse efeito, como também foi
capaz de aumentar a atividade de enzimas antioxidantes (superóxido dismutase e
catalase) no músculo lesionado, e também os níveis do grupo tióis não proteicos (-
SH) nos eritrócitos. Em um segundo trabalho comparamos a combinação de dois
tratamentos (PRP e CWI) muito comumente utilizados no âmbito desportivo. Os níveis
de substâncias reativas ao ácido tiobarbitúrico e diclorofluoresceína oxidada estavam
significativamente aumentados, tanto no tecido muscular esquelético quanto em
eritrócitos, e a combinação de PRP e CWI minimizou esses parâmetros. Além disso,
PRP e CWI combinados foram mais eficazes do que os tratamentos isolados para
aumentar a atividade da catalase, também a relação glutationa reduzida/oxidada e os
níveis do grupo de tióis não proteicos (-SH). Por fim no terceiro trabalho comparamos
os dois métodos de lesão muscular experimental utilizados nos dois trabalhos
anteriores. A contusão e a lesão por estiramento aumentaram os níveis de
marcadores de estresse oxidativo, porém, as lesões mostraram diferenças
significativas entre si nos marcadores de diclorofluoresceína oxidada, atividade da
enzima superóxido dismutase e atividade da enzima N-acetil-beta glucosaminidase
no tecido muscular esquelético. Além disso, as lesões apresentaram diferenças nas
análises comportamentais no teste de caminhada na barra. De modo geral, os
resultados obtidos nesta tese suportam uma nova estratégia para tratamento de
lesões musculares a partir da combinação de PRP e CWI. Além disso, nossos estudos
corroborar para o entendimento da especificidade dessas lesões
musculoesqueléticas, embasando estratégias de tratamento que possam
efetivamente reabilitar essas lesões esportivas, minimizando custos desnecessários
e acelerando o retorno dos atletas à sua prática. | por |
